描述：

ER應激（UPR）反應由三個跨膜傳感器控制：PERK，IRE1和ATF6。這些應激傳感器控制復雜的ER到細胞核信號通路，通過ER膜將信息傳遞給廣泛的基因表達程序。UPR的兩個主要轉錄因子是ATF4和ATF6。ATF4和ATF6都屬于基本的亮氨酸拉鏈ATF / CREB家族，但具有不同的功能UPR。ATF6是II型跨膜ER蛋白，其通過蛋白水解切割而活化?；钚訟TF6易位至細胞核，在那里它與NF-Y和XBP1形成活性同二聚體或二聚體，并與含有ATF / cAMP反應元件（CRE）和/或ER應激元件（ERSE）的啟動子區域結合。ATF4屬于PERK-eIF2-ATF4軸，調節參與氨基酸生物合成和轉運功能的基因的表達，抗氧化應激反應和細胞凋亡。ATF4誘導促存活（早期）和促凋亡（晚期）轉錄程序。長期或的ER應激使用ATF4上調促凋亡蛋白C / EBP同源蛋白（CHOP）。為了區分AT應激中ATF4和ATF6的功能，Signosis開發了ATF4 / ATF6 ELISA試劑盒，用于檢測和比較人，小鼠和大鼠樣品的ATF4和ATF6 DNA結合活性。

原理

TATF6 ELISA試劑盒具有高靈敏度和特異性檢測，操作簡單，優化。用TF共有測序寡核苷酸預先固定96孔（8×12條帶）白板。將核提取物中的活化的TF或全細胞裂解物加入孔中并與寡核苷酸結合。用針對亞基的特異性抗體和HRP綴合的二抗檢測活化的TF。孵育后，洗滌孔以除去未結合標記的抗體。用HRP發光底物進一步檢測該板。發光在微孔板發光計上報告為相對光單位（RLU）。每種特定細胞因子的表達水平與發光強度成正比。

轉錄因子（TF） 是DNA結合蛋白，其在調節基因表達中起重要作用。它們充當傳感器，監測細胞的變化并將信號轉換為基因表達的變化。通常，特定的細胞信號途徑可以激活多個TF，并且特定基因的表達受多個TF的控制。
